The influences of Counting Vials in Estrogen Receptor Assay using Liquid Scintillation Counter


Abstract
Counting vials play an important role in liquid scintillation counting by contributing to background count rate, transmitting light photons and providing a source for photo and recoil electrons upon interaction with gamma rays from an external
standard.
Glass vial is widely used for general counting purpose but it has several shortcomings. Plastic vial is introuduced recently because of overcoming the disadvantages of glass vial.
The author compare the background, total count and value of estrogen receptor using dextran coated charcoal assay between glass and plastic vials in order to evaluate the better one.

@EN 1) the average background in glass vial is 29.7¡¾5.6 CPM and 18.0¡¾3.0 CPM in plastic vial.
2) The average level one value of total count using hydroflur(r) in glass vial is 38295.7¡¾789.0 CPM and 38753.8¡¾413.9 CPM in plastic vial, level two in glass vial is 19249.7¡¾347.4 CPM and 19074.3¡¾735.3 CMP in plastic vial, level thre in
glass
vial
is 10132.4¡¾114.1 CPM and 10319.5¡¾103.3 CPM in plastic vial, level four in glass vial is 6349.3¡¾106.4 CPM and 6379.7¡¾96.5 CPM in plastic vial. Level five in glass vial is 3283.4¡¾47.4 CPM and 3387.0¡¾138.5¡¾CPM in plastic vial, level six in
glass
vial is 2070.2¡¾40.2 CPM and 2088.0¡¾46.6CPM in plastic vial. No statistical difference in total count using hydroflur(r) between each vials except level III and V is observed.
3) The average estrogen receptor value in glass vial is 78.5¡¾29.7 femtomole/mg cytosol protein and 74.2¡¾26.8 femtomole/mg cytosol protein in plastic vial.
With the above results, the author conclude that plastic vial is the alternative to glass vial irrestrogen receptor assay using liquid scintillation counter.
